Mechanism of Action
Senescent cells upregulate FOXO4, which sequesters p53 in the nucleus and prevents it from triggering apoptosis — allowing senescent cells to persist and secrete the pro-inflammatory SASP (senescence-associated secretory phenotype). FOX04-DRI is a D-retro-inverso peptide that competitively disrupts the FOXO4–p53 interaction, freeing p53 to activate its apoptotic programme. Because healthy cells do not rely on this FOXO4–p53 axis for survival, FOX04-DRI selectively eliminates senescent cells without cytotoxicity to normal tissue. Research models show clearance of p21-positive senescent cells in liver, kidney, and intestinal tissue following administration.
